Gene Overexpression Reagent Checklist and Vector Selection Guide
Miaoling Biotechnology offers a range of in-stock expression vectors for mammalian cells, E. coli, yeast, plants, and other experimental systems to support a variety of research applications.
Introduction
Gene overexpression involves cloning a target gene’s coding sequence into a suitable expression vector and introducing it into a host cell or organism through methods such as transfection or viral transduction. The goal is to increase expression at the mRNA level, the protein level, or both. This technique is widely used in gene function studies, signaling pathway analysis, recombinant protein production, and gene therapy research.
A typical gene overexpression experiment requires several core reagents and consumables: an overexpression plasmid, a host cell or strain, transfection or transformation reagents, selection antibiotics when needed, and nucleic acid or protein detection reagents.

Reagent Checklist and Vector Selection Guide
Selecting an Overexpression Vector
Transient Overexpression in Mammalian Cells: pCMV Series
Typical applications: Short-term expression, often assessed over approximately 3–7 days, without establishing a stable cell line. This approach is useful for rapid evaluation of gene function.
Recommended in-stock products from Miaoling Biotechnology
Untagged constructs
- P72762 — pCMV-CHP2(human)-Neo: Suitable for applications where a fusion tag could interfere with downstream assays.
- P73081 — pUC57-Dlat(Rat): A plasmid containing the target gene sequence that may serve as a DNA template, subject to confirmation of its construct design. Note that a pUC57 cloning vector should not automatically be treated as a mammalian expression vector.
Single-tagged constructs for protein detection by Western blotting
- P72865 — pCMV-SEL1L3(human)-3×FLAG-Neo
- P74710 — pCMV-YME1L1(human)-3×HA-Neo
- P73652 — pCMV-Sox4(mouse)-3×FLAG-Neo
- P73009 — pCMV-Egln1(mouse)-3×HA-Neo
Fluorescently tagged constructs for expression tracking and localization studies
- P72764 — pCMV-ABCG2(human)-GFP-Neo

Construct containing a WPRE regulatory element
- P73007 — pCMV-T7-E2F8(human)-3×FLAG-WPRE-Neo
WPRE can enhance transgene expression in certain vector contexts, but the magnitude and direction of its effect depend on the construct and experimental system. It should not be assumed to increase protein abundance in every application.
Suggested transfection reagents: Lipid-based transfection reagents, such as Lipofectamine 2000 or Lipofectamine 3000, or suitable cationic polymer-based reagents. Optimize the reagent-to-DNA ratio for the specific cell type and plasmid.
Stable Overexpression in Mammalian Cells: pLV3 Lentiviral Series
Typical applications: Longer-term expression and delivery to cell types that may be difficult to transfect, including some primary cells, suspension cells, and stem cells.

Suggested lentiviral packaging components: A compatible packaging system, commonly including psPAX2 and pMD2.G, together with an appropriate packaging cell line such as 293T cells. Confirm compatibility with the transfer vector and packaging system before use.
For viral preparations, follow the recommended storage conditions and minimize repeated freeze–thaw cycles to help preserve functional titer. Stable expression should be confirmed after transduction and selection rather than inferred from the presence of a selectable marker alone.
Recombinant Protein Expression in Prokaryotes: pT7 and pET Series
Typical applications: Recombinant protein production in E. coli when purified protein is needed for in vitro experiments, biochemical assays, or antibody development.
Recommended in-stock products from Miaoling Biotechnology
- P74614 — pT7-6×His-TXN(human): Contains a 6×His tag for affinity purification using a compatible nickel-based resin.

The required materials depend on the vector architecture and the intended delivery method. For example, a zebrafish construct used for transgenesis may require a different workflow from one used for mRNA injection. Verify the product configuration and recommended application before selecting supporting reagents.
Choosing Selection Antibiotics
The appropriate selection antibiotic depends on the resistance marker encoded by the vector and the host system. The following concentrations are general starting references, not universal recommendations.
|
Vector Resistance Marker |
Common Antibiotic |
Example Starting Range or Condition |
|
Neo/G418 in mammalian cells |
G418 sulfate |
200–1,000 μg/mL |
|
Puro in mammalian cells |
Puromycin |
1–10 μg/mL |
|
Blast in mammalian cells |
Blasticidin S |
5–10 μg/mL |
|
Hyg in mammalian cells |
Hygromycin B |
100–500 μg/mL |
|
Amp in bacteria |
Ampicillin |
50–100 μg/mL |
|
Kan in bacteria |
Kanamycin |
50 μg/mL as an example starting condition |
Important: The optimal concentration varies with cell type or bacterial strain, culture conditions, and the resistance construct. For mammalian stable-cell-line selection, perform a kill-curve experiment using untransfected cells before selecting a working concentration. Bacterial selection and plant transformation require their own system-specific conditions.
Miaoling’s pCMV-series products often carry Neo resistance markers, while many pLV3-series products carry Puro markers. Always verify the marker listed for the individual product rather than assuming that every vector in a series uses the same selection system.

Select the assay according to the intended endpoint. RT-qPCR measures transcript abundance, whereas Western blotting and other protein-based methods assess protein abundance, localization, or related properties. Neither mRNA nor protein abundance alone necessarily demonstrates biological function.
Direct observation of fluorescent reporters
If the expression construct contains a fluorescent reporter such as GFP, CopGFP, EYFP, or mCherry, reporter fluorescence can often be observed approximately 24–72 hours after transient transfection, depending on the construct and cell type. Additional staining is generally unnecessary to detect the fluorescent reporter itself.
Miaoling products such as P72764, P72635, and P73078 contain fluorescent tags and may be suitable for fluorescence microscopy or live-cell imaging, subject to confirmation of the construct design and experimental compatibility.
Fluorescence indicates reporter expression, not necessarily correct localization or biological activity of the target protein. For fluorescent fusion constructs, confirm that the tag does not interfere with the protein’s function or trafficking, and use an independent assay when necessary.
